简介

癌症是以基因突变导致细胞异常和失控生长为特征的一系列疾病。驱动基因(Cancer Driver Gene, CDG)是指对肿瘤进展有重大影响的基因。根据在癌症进展中的作用,驱动基因可分为两大类:原癌基因(Oncogenes)和抑癌基因(Tumor Suppressor Genes)。癌基因成瘾(oncogene addiction)是指某些肿瘤维持其恶性生物学表型依赖于某个或某些活化癌基因的现象,这些癌基因也称为驱动癌基因(driver oncogenes)。

例如,EGFR基因突变是肺癌中一种常见的突变,它使得EGFR受体持续处于激活状态,信号传递过程异常,导致细胞长期无节制地增殖和生长,成为肺癌细胞的驱动基因之一。癌细胞需要驱动癌基因持续发挥功能,而正常细胞则不需要。因此,以癌基因为治疗靶点,可以使靶向药物特异性地杀伤肿瘤细胞,而不损伤正常细胞。

Martínez-Jiménez等提出IntOGen流程,应用于66种癌症类型28,000多例肿瘤的体细胞突变,揭示了568个癌症基因,并指出了它们的肿瘤发生机制。这些癌症驱动基因的鉴定促进了靶向抗癌疗法模式的发展,更广泛地说,促进了对预后和治疗反应基因组生物标志物的研究。
