简介

趋化因子(Chemokines)或称趋化细胞因子,是由细胞分泌的一系列小细胞因子或信号蛋白,可诱导白细胞以及包括内皮细胞和上皮细胞在内的其他类型细胞定向移动。除了在激活宿主免疫反应中发挥重要作用外,趋化因子对生物过程(包括形态形成和伤口愈合)以及癌症等疾病的发病机制也很重要。

趋化因子主要分为四个亚家族:CXC、CC、CX3C和C。一些趋化因子被认为具有促炎作用,可在免疫反应期间被诱导,将免疫系统的细胞招募到感染部位,这些趋化因子包括CCL14、CCL19、CCL20、CCL21、CCL25、CCL27、CXCL12和CXCL13。这种分类并不严格,例如,CCL20也可以作为促炎趋化因子。其他趋化因子则被认为是稳态趋化因子,在组织维持或发育的正常过程中参与控制细胞迁移。这些趋化因子在病理条件下形成(在促炎性刺激下,如IL-1、TNF-α、LPS或病毒),并积极参与炎症反应,将免疫细胞吸引到炎症部位,如CXCL-8、CCL2、CCL3、CCL4、CCL5、CCL11、CXCL10。

趋化因子受体是一种G蛋白偶联受体,含有7个跨膜结构域,存在于白细胞表面。趋化因子受体与G蛋白结合后可传递细胞信号。受体激活后,G 蛋白的α-和β-γ-亚基解离,激活多种下游通路,导致细胞极化和肌动蛋白重组。小 GTP 酶的各种成员都参与了这一过程。一氧化氮的诱导和活性氧的产生也受到趋化因子信号通过钙动员和二酰甘油产生的调节。
